OF THE DISCLOSURE Cryopreservation of endothelial cell monolayers is one of the major challenges in the cryopreservation of complex tissues. Human umbilical vein endothelial cells (HUVECs) in suspension are available commercially and recently their post-thaw cell membrane integrity was significantly improved by cryopreservation in 5% dimethyl sulfoxide (DMSO) and 6% hydroxyethyl starch (HES). Here, we show that a combination of different factors contribute to significant progress in cryopreservation of cell monolayers. The addition of 2% chondroitin sulfate to 5% DMSO and 6% HES and cooling at 0.2 or 1 °C/min led to high membrane integrity (97.3 ± 3.2%) immediately after thaw when HUVECs were cultured on a substrate with a coefficient of thermal expansion similar to that of ice. The optimized cryopreservation protocol was applied to monolayers of primary porcine corneal endothelial cells, and resulted in high post-thaw viability (95.9 ± 3.7% membrane integrity) with metabolic activity 12 hours post-thaw comparable to unfrozen control.

Smart pH-sensing microencapsulation system programmed to actively respond to environmental pH change has long been considered a key technology in pharmaceutical industries and food sciences. Maintenance of both stability of active materials in harsh gastrointestinal (GI) tract environments and release at target site-specific pH is a major obstacle in oral drug/vaccine development. In this study, we developed a first-of-its-kind drug delivery system with pH-responsive macropores. Opening and closing of these macropores depend on pH to meet the critical requirements of intestine-targeted drug delivery system, providing stability in stomach and rapid release in intestine. To this end, we developed pored hollow microparticles (MPs) made of anionic copolymer using an Oil-in-Water (O/W) emulsion method. We observed that freeze-drying of MPs induced closure of macropores on their surface, probably due to the decrease of glass transition temperature via surfactant incorporation and/or physicochemical characteristics of polymer-water and polymer-polymer interaction during dehydration process. The closing and opening behavior of the sealed macropore on MPs in response to pH change were confirmed after being subjected to simulated gastric pH (2-hr incubation at pH 2.0) and intestinal pH (4-hr incubation at pH 7.1), respectively. Time dependent release behavior of ingredients through pH-sensing pores was tested using two different types of model drugs (sulforhodamine b and 100 nm sized fluorescent nanoparticles). The intact, closed structure of the dye-encapsulated macropore was maintained in simulated gastric environment. Subsequent exposure of the same sample to simulated intestine environment induced pore opening, which caused release of the ingredients from MPs. To demonstrate the applicability of our microencapsulation system for active biomolecules, lactase enzymes were encapsulated in MPs. Theses MPs displayed more than 15 times higher protections than commercial lactase formulations, indicating the protective activity of the MP system against harsh GI conditions. This study showed that taking mutual advantages of solid and hollow architectures, microparticles with a pH-responsive macropore exhibited easy solvent-free loading and protection of ingredients in unfavorable pH by closing pores, and triggered their release in targeted pH by opening pores. Ultimately, our MP system with a pH-sensing macropore strives to usher a new area of research in smart drug delivery systems and advance the current oral drug delivery technology by solving major challenges in targeted delivery of pH- sensitive therapeutic agents. 67
