Abstract

Provided are compositions and methods for promoting tolerance to auto-immune antigens. In general the compositions include quantum dots (QDs) that are in association with auto-immune peptide antigens. It is shown that QDs can be used to generate immunological tolerance by controlling the density of self-antigen on QDs. Peptide-QDs rapidly concentrate in draining lymph nodes, and co-localize with macrophages expressing scavenger receptors involved in tolerance. Treatment with peptide-QDs reduces disease incidence 10-fold. The degree of tolerance and the underlying expansion of regulatory T cells correlates with the density of myelin molecules presented on QDs such that higher numbers of tolerogenic particles displaying lower levels of self-peptide are more effective for inducing tolerance than fewer particles each displaying higher densities of peptide. The disclosure is therefore relevant to promoting tolerance to antigens that are involved in a variety of autoimmune disorders.

Abstract

An oscillatory apparatus and methods of utilizing the same. In one embodiment, the apparatus comprises a force sensor having a proof mass, with one or more sensing electron tunneling electrodes disposed thereon, and a frame comprising one or more reference electron tunneling electrodes. Conductive plates disposed on the sensor base and capping wafers induce oscillations of the proof mass. The sensing and the reference electrode pairs are disposed in a face-to-face configuration, thus forming a digital switch characterized by one or more closed states. In the closed state, the switch generates triggering events, thereby enabling the sensing apparatus to generate a digital output indicative of the mass position. The time period between consecutive trigger events is used to obtain mass deflection due to external forcing. Time separation between the triggering events is based on the physical dimensions established during fabrication, thus not requiring ongoing sensor calibration.

Abstract

A method for manufacturing or preparing thin-film stacks that exhibit moderate, finite, stress-dependent resistance and which can be incorporated into a transduction mechanism that enables simple, effective signal to be read out from a micro- or nano-mechanical structure. As the structure is driven, the resistance of the intermediate layers is modulated in tandem with the motion, and with suitable dc-bias, the motion is directly converted into detectable voltage. In general, detecting signal from MEMS or NEMS devices is difficult, especially using a method that is able to be integrated with standard electronics. The thin-film manufacturing or preparation technique described herein is therefore a technical advance in the field of MEMS/NEMS that could enable new applications as well as the ability to easily develop CMOS-MEMS integrated fabrication techniques. Also disclosed are: (i) transducers where current flows across a piezo layer from one major surface to the opposite major surface; and (ii) methods of making a transducer the resistivity of a piezoresistive layer is decreased and/or the gauge factor of a piezoresistive layer is increased.
