Abstract

Cell sorting methods that improve sorting efficiency and productivity by elevating sorting pressures and incorporate certain steps to help the cells better survive such elevated pressures. In the case of sperm, sorting the steps of standardizing sperm samples, staining sperm samples in a single step, calibrating a flow cytometer to place sperm in the leading edge of droplets, and changing a catch fluid distance may be incorporated individually, or in combination to help sperm better survive the sex sorting process.

Abstract

The present invention generally relates to compositions and methods for the handling of processed sperm populations including samples that are freshly collected, those transported as fresh samples, as well as samples that are frozen and thawed, those sorted into one or more subpopulations, and those that are otherwise processed or handled that impose trauma on the sperm cell. Such trauma can reduce the motility, fertility, viability and overall integrity of the sperm and reduce the sperm's ability to fertilize an egg, grow into a health embryo and produce a healthy offspring. The present invention relates to novel compounds that can be added to the sperm cell sample to reduce the traumatic effects of physical stress during mild as well as extensive sperm cell processing, methods of using the compounds in standard sperm processing procedures, the end products made from these methods including sperm and embryos, as well as methods of using those end products in assisted reproductive biology techniques in animals.

Abstract

Generally, compositions and methods for handling processed sperm populations including samples that are freshly collected, transported as fresh samples, as well as samples that are frozen and thawed, those sorted into one or more subpopulations, and those that are otherwise processed or handled that impose trauma on the sperm cell. Such trauma can reduce the motility, fertility, viability and overall integrity of the sperm and reduce the sperm's ability to fertilize an egg, grow into a healthy embryo and produce a healthy offspring. The novel compounds described can be added to the sperm cell sample to reduce the traumatic effects of physical stress during mild as well as extensive sperm cell processing, methods of using the compounds in standard sperm processing procedures, the end products made from these methods including sperm and embryos, as well as methods of using those end products in assisted reproductive biology techniques in animals.

Abstract

A method of liquid nitrogen surface vitrification requiring an embryo washed in a rinsing medium, then incubated in a base medium and incubated in a hold medium before being washed in a vitrification medium and produced into a vitrification droplet (270). For forming the droplet, vitrification medium (210), an intermediary fluid such as air, followed by vitrification medium containing at least one embryo (250) are aspirated into the channel. The vitrification droplet consequently can contain an air bubble (220). The vitrification droplet can be produced from an instrument with a channel and dropped directly into liquid phase nitrogen producing a vitrified droplet. The vitrified droplet can be stored in cryo-vessels, and warmed for revitalization of biological function of vitrified biological cell mass or tissues, such as oocytes and/or embryos.

Abstract

A method of sex sorting sperm is disclosed. The sperm may be stained with a DNA selective fluorescent dye, which fluoresces when excited, a dead quenching dye, which selectively quenches fluorescence emitted by the DNA selective fluorescent dye within the membrane of compromised sperm, and a split enhancing dye. The stained sperm may then sorted into one or more gender enriched subpopulations of viable sperm.

Abstract

Staining mixtures comprising viable spermatozoa, a composition which regulates oxidation/reduction reactions intracellularly or extracellularly, and a DNA selective dye are disclosed. The cells contained in such suspensions tend to have a greater capacity for enduring the various process steps typically associated with the sorting of sperm cells into gender enriched populations, thereby resulting in post-sort compositions with an increased number of viable or motile sperm. Processes for staining sperm cells comprising the formation of a staining mixture are also disclosed.

Abstract

Processes of storing sorted and unsorted spermatozoa, in the form of a sperm dispersion, having reduced motility relative to endogenous ejaculated sperm are disclosed. The immotile spermatozoa of the dispersion tend to have a greater capacity for enduring the rigors associated with storage, transportation, and fertilization procedures. Processes for providing such sperm dispersions, for inseminating a female mammal with such sperm dispersions, as well as compositions and combinations comprising the sperm dispersions, are also disclosed.

Abstract

A multichannel system for classifying particles according to one or more characteristics of the particles includes a plurality of flow cytometry units. Each flow cytometry unit is operable to classify particles in a mixture of particles by interrogating a stream of fluid containing particles using a beam of electromagnetic radiation. Each flow cytometry unit has a sensor operable to generate a time-varying output signal indicative of at least one characteristic of the particles in the stream of fluid as the stream of fluid is interrogated by the beam of electromagnetic radiation. The flow cytometry units share an integrated platform and perhaps even a common processor for receiving and processing information from the units. The common processor is programmed to receive the time-varying output signals from the flow cytometry units substantially continuously and to process the output signals.

Abstract

Sperm cells are stained according to processes that involve the combining of sperm cells with a fluorescent DNA selective dye at an elevated temperature in excess of about 40° C. The methods allow for a decreased staining time. The cells may thereafter be efficiently sorted according to common separation methods, including flow cytometry.
